چکیده

Viruses have proven to be of major importance in medical science both as infectious agents and as models for molecular studies. Animal viruses containing deoxyribonucleic acid (DNA) genomes, in particular, provide simple model systems that can be used to address many complex biological problems. All animal DNA viruses have small genomes, relative to the host cells they infect, that can be easily manipulated to examine the mechanisms of replication and

A codon-shuffling method to prevent reversion during production of replication-defective herpesvirus stocks: Implications for herpesvirus vaccines

Herpesviruses establish life-long chronic infections that place infected hosts at risk for severe disease. Herpesvirus genomes readily undergo homologous recombination (HR) during productive replication, often leading to wild-type (WT) reversion during complementation of replication-defective and attenuated viruses via HR with the helper gene provided in trans. To overcome this barrier, we deve...

The strain-specific cis-acting element of beet curly top geminivirus DNA replication maps to the directly repeated motif of the ori.

Strains of beet curly top geminivirus (BCTV) possess distinct cis- and trans-acting replication specificity elements which are not separately interchangeable among strains. Analysis of the replication competency of chimeric BCTV genomes, in which portions of the origin of DNA replication (ori) were derived from heterologous BCTV strains, have permitted identification of an essential cis-acting ...

Analysis of Essential Viral Gene Functions after Highly Efficient Adenofection of Cells with Cloned Human Cytomegalovirus Genomes

Human cytomegalovirus (HCMV) has a large 240 kb genome that may encode more than 700 gene products with many of them remaining uncharacterized. Mutagenesis of bacterial artificial chromosome (BAC)-cloned CMV genomes has greatly facilitated the analysis of viral gene functions. However, the roles of essential proteins often remain particularly elusive because their investigation requires the cum...

A conditionally replicating HIV-1 vector interferes with wild-type HIV-1 replication and spread.

Defective-interfering viruses are known to modulate virus pathogenicity. We describe conditionally replicating HIV-1 (crHIV) vectors that interfere with wild-type HIV-1 (wt-HIV) replication and spread. crHIV vectors are defective-interfering HIV genomes that do not encode viral proteins and replicate only in the presence of wt-HIV helper virus. In cells that contain both wt-HIV and crHIV genome...

9 Protein - Primed Replication of Bacteriophage 29 DNA

Unlike circular genomes in which the conventional replication fork machinery can accomplish the copy of the complete molecule, the requirement of a DNA/RNA molecule to prime DNA synthesis imposes replication strategies to avoid the loss of genetic information contained at the very end of the lagging strand in linear chromosomes since DNA polymerases are unable to start de novo DNA synthesis. Th...
